Bacterial DNA Extraction using InstaGene® Matrix

In spite of using Phenol and Chloroform to extract bacterial DNA template, you can use the InstaGene matrix to be more efficient. This DNA extraction method is very easy, fast, and practical. Here is the procedure:

  1. Preparing 1.5 ml sterile tube
  2. Filling the tube with 250 micro liter sterile water (for DNA purpose), then dissolving 1 to 2 ose of pure bacteria isolate until the solution look turbid
  3. Centrifuge the tube at 10000-12000 rpm at 4 Celcius degree for 1 minute.
  4. Taking the pellet then adding 50 micro liter of InstaGene matrix.
  5. Incubate the tube in 56 Celcius degree water-bath for 45 minutes.
  6. Afterwards, incubate the tube in 100 Celcius degree water-bath for 8 minutes.
  7. Centrifuge the tube at 10000 to 12000 rpm at 4 Celcius degree for 2 to 3 minutes.
  8. Supernatant will contain the bacterial DNA template.
